OBJECTIVES
  • To understand the relationships between genetic background and diet/lifestyle, as well as to comprehend the details of the mechanisms underlying insulin-related disturbances within the endothelium and Type 2 diabetes (T2D) onset.
  • To understand the positive or negative effects of dietary supplementation in relation to diabetes-induced metabolic disorders.
  • The study will focus mainly on the effects of resveratrol, which activates sirtuin 1, a molecular target for the treatment of metabolic disorders, on commercially available human endothelial cells chronically treated with high-glucose.
 
 
 
 
EXPECTED RESULTS

The project is aimed at revealing the molecular details of the possible bio-effects of the activation of SIRT1-dependent pathway in response to high-glucose-induced cellular subtoxicity in human umbilical vein endothelial cells (HUVECs).

 
 
 
 
PLANNED SECONDMENT

First secondment: to acquire advanced technological skills in protein expression analysis and gene profiling under the supervision of Prof. Antonio Giordano, at Temple University, Philadelphia, PA, USA.

 

Second secondment: chemical design and validation of alternative SIRT1 activators under the supersivion of Prof. Marcello Allegretti, at Dompè Farmaceutici Spa (a Pharma Company located in L’Aquila).
